This is a known behavior with the Act! 2010 V12.1 program, the Salutation field will not populate until a 'Save' is issued for the contact. The 'Save' can be prompted through several actions, the easiest of which is to click on the 'Save Changes' icon on the toolbar (or use the key combination 'CTRL + S').
This has been reported to the Development team.
