We operate with act 2007 and are upgrading to 2010 in January, however we are also looking into the premium version.
1) Does the premium version of 2010 allow you to attach specific layouts to specific users.
For example, accounting does not need to see marketing tabs
2) Can one color the tab labels?

ACT! Premium doesn't provide any further options for layout management based on user identities; it's just the same as in previous versions of Standard or Premium edition. You can certainly create a range of different layouts that are selectable from a drop-down list (as ever), and each users layout would default to the last-used layout on his/her machine.
If you want to take it further than this, there are add-ons that may be of interest:
layout switch allows you to automate the presentation of a specified layout based upon the contents of any referenced field
Durkin Toolkit includes some components that enable you to customise tabs in layouts beyond what's available within the base product

The default last used layout (which I never new) plus field level security in premium should do the job for now.
The addons look like great tool, will review in the future.
