I have used the scan for duplicate feature in the past. I'm now getting an error message and it won't perform a scan.
I went to Tools, Preferences, General Tab and moved down to Duplicate Checking. The enable duplicate checking box is grayed out and I'm not able to choose it.
How would I correct this problem?

Are you using a remote database? The option is disabled in remotes, it would need to be set in the main/publisher database and then synchronization will pick up the settings.
