A client had a computer with 2 Hard Drives: C and D. Act was on the D drive and it failed. Act is now on the C drive but there are no layouts available because act is looking for the D drive which no longer exists.
How do I tell act where to find the layouts folder on the C drive? I reset the preferences without success.

I'm assuming that the layouts are where they are supposed to be, but not showing on the main screen.
Not sure if this will work, but try opening the layout editor and open another layout. Navigate to the default location on the C drive and see if it is there. If so, open and then save as. See if that will solve the problem.
Otherwise, I bet you get to dive into the registry.
